    Research on the Phenotypic Characterization of Mrsa Strains Isolated from Animals

    Keywords: chromogen, methicillin, MRSA, resistanceIntroduction: Currently, both in staphylococci isolated from animals with different diseases, as well as in humans, the MRSA strains (Methicillin Resistant S. aureus) are monitored, as the methicillin resistance is associated with the resistance to other antibiotic groups.Methicillin resistance is encoded by mec staphylococcal chromosomal cassettes (SCCmec), which are islands of resistance. These strains can be identified by molecular biology tests and tests that reveal several phenotypic characteristics.The research was made in order to characterize and identify phenotypically the MRSA staphylococci strains isolated from animals.Materials and Methods: Researches were made on 240 coagulase positive and coagulase negative strains of staphylococci. Mannitol fermentation was tested on Champan medium, free coagulase was revealed on Baird-Parker medium and to identify S. aureus subsp. aureus was used the chromogenic medium Chromatic Staph.Methicillin-resistant strains were detected by disc diffusion method, using biodiscs with methicillin, oxacillin and cefoxitin. Also, to identify the MRSA strains, was used the chromogenic medium Chromatic MRSA.Results: The isolates were positive to mannitol and produced complete haemolysis or were unhaemolytic. A total of 44 strains produced free coagulase on Baird-Parker medium, considered coagulase positive strains, while 196 were coagulase negative strains.The isolates conducted differently to methicillin: 22,08% of strains were resistant, 51,25% of strains were susceptible and 26,66% had intermediate resistance, while the resistant strains to oxacillin were 42,91%.The increased frequency of methicillin-resistant strains of staphylococci and, particularly, MRSA strains, determined using the cefoxitin disk diffusion test, which is more reliable than methicillin and oxacillin.On the MRSA chromogenic medium, the methicillin-resistant strains of staphylococci formed colonies with pigment from mauve to orange mauve.Conclusion: The obtained results by disk diffusion test on resistance patterns to 3 beta-lactams, resistant to penicillinase, indicated a different frequency of the resistant strains to these antibiotics.Cefoxitin disk diffusion test revealed a frequency of 2,51% of resistant strains, that can be considered MRSA strains

    Comparative Study on Quality Parameters of Royal Jelly, Apilarnil and Queen Bee Larvae Triturate

    Given their beneficial effects in terms of health, the natural products, especially beehive products, have drawn the attention of consumers since long time ago. In order to guarantee the quality of these products on the market, their chemical composition needs to be analyzed. Thus, this current research had as objective the establishment of quality parameters for beehive brood food derived products: apilarnil and queen bee larvae triturate. These two products were compared with royal jelly which is the basis of brood food in the first 3 days of larval stage. The carbohydrates were determined by HPLC-IR and allowed the identification of seven carbohydrate compounds, predominantly glucose, fructose and sucrose. The lipid profile was analyzed by the Soxhlet method. The total protein content was determined by the Kjeldahl method. Free amino acids were analyzed by LC-MS. A total of 31 amino acids were identified of which nine are essential amino acids for humans.Â

    Do Polymorphisms of the TERT, GSTM1, and GSTT1 Genes Increase Laryngeal Cancer Susceptibility in Smokers of Romanian Descent?

    Background and Objectives: The aim of this study was to investigate the association between smoking status and single-nucleotide polymorphism in candidate genes that had a known association with smoking-related tumors in previous studies and to explore their link to laryngeal cancer risk in a population of northern Romanian descent. The genes selected have key functions in xenobiotic metabolism (GSTs: the glutathione S-transferases family: GSTM1 and GSTT1) and chromosomal management (TERT). Materials and Methods: The genotype frequencies of TERTRs2736100 and the GST subfamilies (GSTM1 and GSTT1) were determined using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique. The relationship between the polymorphisms and the risk of laryngeal cancer was analyzed in a retrospective case–control study of 92 laryngeal cancer cases and 101 controls, all of whom were smokers. Results: Subjects presenting the GSTT1-null variant had a two-fold increase in risk (OR = 2.05, 95% CI = 1.07–3.95, p = 0.02). While no individual risk was observed for the TERTRs2736100 polymorphism, stratification based on gender revealed a nine-fold increase in risk for carriers of the “C” allele in the heterozygote variant who were male (OR = 9, 65% CI = 3.51–26.51, p = 0.0000). Conclusions: The results showed that the GSTT1-null genotype and the mutant heterozygote variant of TERTRs2736100 genes may play a significant role in laryngeal cancer susceptibility in subjects of northern Romanian descent. There may be no association between the susceptibility to laryngeal carcinoma and the GSTM1 polymorphism. The results could not confirm the carcinogenic influence smoking has on laryngeal cancer development for the studied polymorphisms

    Assessment on the influence of TLR4 and DNA repair genes in laryngeal cancer susceptibility: a selective examination in a Romanian case control study

    Background: Tumor characterization through the study of molecular biology has become an invaluable tool in understanding cancer development and evolution due to its relationship with chromosomal mutations, alterations or aberrations. The purpose of this study was to investigate the involvement of genes such as TLR-4 and DNA repair pathways (XRCC1 and XPD) in laryngeal cancer susceptibility in a Romanian population. Method: We performed a case-control study on 157 laryngeal cancer patients and 101 healthy controls. Genetic testing was carried out using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism. Results: We identified the Gln allele of the XPDLys751Gln polymorphism as an individual risk factor in laryngeal cancer development (Gln vs Lys, adjusted OR=1.65, 95%CI=1.13–2.40, P=0.008). Subjects with the mutant homozygote variant (Gln/Gln) had a two fold increase in cancer risk (adjusted OR=2.18, 95%CI=1.06–4.47, p=0.028) when compared to the reference wild type genotype (Lys/Lys). Stratification by sex and age, identified males under 62 years as the most susceptible group with an almost three fold risk (adjusted OR=2.94, 95%CI=1.31–6.59, p=0.007) for the dominant model (Lys/Gln+Gln/Gln). No associations were found for TLR-4Thr399Ile, XRCC1Arg194Trp and XRCC1Arg399Gln. Conclusion: The results of the study show that the XPDLys751Gln polymorphism may be among other independent risk factors for developing laryngeal cancer where as TLR-4Thr399Ile, XRCC1Arg194Trp and XRCC1 Arg399Gln show no such association. However, we consider the relative small number of the subjects selected for this analyses a possible limitation towards the real influence the obtain results may pertain in laryngeal cancer evolution

    Diagnostic challenge – West Nile encephalitis mimicking stroke – case report

    West Nile virus (WNV) is widely distributed around the world. The WNV is maintained in nature by a cycle involving mosquitoes and birds, which are presumed to be the most important amplifying hosts. We present the case of a 75 years old male patient admitted to the neurology department for several symptoms that could be attributed to a stroke. The symptomatology had an acute onset. The computed tomography and the magnetic resonance imaging revealed lacunar stroke in right semioval center. The final diagnosis was established by the lumbar punction: West Nile virus was highlighted in the cerebrospinal fluid,clinical evolution was favorable, the patient was discharge with minimal neurologic deficits
